Egr-1 Enhances Drug Resistance of Breast Cancer Cells by MDR1 Dependence

چکیده

Paclitaxel (PTX) is one of the most effective drugs used in treatment breast cancer. Nonetheless, appearance MDR1 (multidrug resistance 1) tumor cells has become a significant hindrance for efficacious chemotherapy. In this study, we show that expression level Egr-1 (early growth response gene-1) cancer tissues (from paclitaxel chemotherapy failure patients) and MCF-7/PTX (the cell line was resistant to paclitaxel) increased. Cell proliferation assay apoptosis revealed could promote inhibit MCF-7/PTX. Mechanistic studies indicated bind proximal promoter enhance transcription. These findings indicate induced accumulation upregulated MDR1, thereby inducing drug Our results suggest novel pathway by which induces expression, possibly illuminating potential target prevention MDR1-mediated resistance.

Nanolipoparticles-mediated MDR1 siRNA delivery reduces doxorubicin resistance in breast cancer cells and silences MDR1 expression in xenograft model of human breast cancer

Objective(s): P-glycoprotein (P-gp) is an efflux protein, the overexpression of which has been associated with multidrug resistance in various cancers. Although siRNA delivery to reverse P-gp expression may be promising for sensitizing of tumor cells to cytotoxic drugs, the therapeutic use of siRNA requires effective carriers that can deliver siRNA intracellularly with minimal toxicity on targe...

Down regulation of CIAPIN1 reverses multidrug resistance in human breast cancer cells by inhibiting MDR1.

Cytokine-induced apoptosis inhibitor 1 (CIAPIN1), initially named anamorsin, a newly indentified antiapoptotic molecule is a downstream effector of the receptor tyrosine kinase-Ras signaling pathway. Current study has revealed that CIAPIN1 may have wide and important functions, especially due to its close correlations with malignant tumors. However whether or not it is involved in the multi-dru...

Cancer Stem Cells and Multiple Drug Resistance in Breast Cancer*

Million of new cancer patients are diagnosed each year and over half of these patients die from this disease. As the second leading cause of cancer deaths, breast cancer in estimated to be diagnosed in over one million people worldwide and to cause more than 400,000 deaths each year. Chemotherapy is part of a successful treatment to many cases; however, the development of multidrug resistance (...

MMP1 expression is activated by Slug and enhances multi-drug resistance (MDR) in breast cancer

High matrix metalloproteinase 1 (MMP1) expression is associated with enhanced breast cancer growth and metastasis and also might predict poor prognosis. In this study, we further investigated the functional role of MMP1 and how it is upregulated in multi-drug resistant (MDR) breast cancer cells. By retrieving microarray data in GEO datasets and the survival data in the Kaplan Meier plotter, we ...
